چکیده

Microcalorimetric titrations of calmodulin with Ca" and trifluoperazine (TFP) at various molar ratios have been carried out at 25 "C and at pH 7.0. Ca2+ binding to calmodulin produces heat ( A H ) in the presence of TFP, while heat is absorbed in the absence of TFP. The total heat produced by Ca2+ binding to all four sites is increased at increasing TFP-to-calmodulin ratios, attaining a plateau at about 7. These results indicate that at the higher ratios, the enthalpy changes ( A l l ) associated with Ca2+ binding are affected by TFP molecules bound at both highand low-affinity sites. In addition, the Ca2+ binding reaction of the calmodulin-TFP complex is driven solely by a favorable enthalpy change of -27 kJ/mol of site; the entropy change (As) is -35 J/ mol/K. These thermodynamic changes are opposite to those for TFP-free calmodulin and distinctly different from other Ca2+ binding proteins such as skeletal and cardiac troponin C and parvalbumin, where the reaction is driven by favorable changes of entropy as well as enthalpy.

Regulation of phosphorylase kinase in rat ventricular myocardium. Role of calmodulin.

Conversion of phosphorylase b to a which is catalyzed by the enzyme phosphorylase kinase is known to require Ca++. Trifluoperazine, an inhibitor of calmodulin-dependent enzymes, was utilized in the present study to clarify the role in vivo of calcium-calmodulin regulation of phosphorylase kinase. Twenty-minute preperfusion of isolated rat ventricles with 10(-5) M trifluoperazine had no effect o...

Effects of trifluoperazine on function and structure of toad urinary bladder. Role of calmodulin vasopressin-stimulation of water permeability.

Calcium ion plays a major regulatory role in many hormone-stimulated systems. To determine the site of calcium's action in the toad urinary bladder, we examined the effect of trifluoperazine, a compound that binds specifically to the calcium binding protein, calmodulin, and thereby prevents activation of enzymes by the calcium- calmodulin complex. 10 microM trifluoperazine inhibited vasopressin...

A novel calcium binding protein in Mycobacterium tuberculosis--potential target for trifluoperazine.

Phenothiazines have been reported for anti-mycobacterial activity by inhibiting calcium binding proteins, potassium transport processes of phagolysosomes, NADH dependent oxygen consumption by M. tuberculosis membranes and DNA, and lipid synthesis of the bacterium. Thioridazine (TZ), chloropromazine (CPZ) and trifluoperazine (TFP) belong to the class of phenothiazines widely used as neuroleptic ...

Effects of trifluoperazine on calcium binding by calmodulin. A microcalorimetric study.

Microcalorimetric titrations of calmodulin with Ca2+ and trifluoperazine (TFP) at various molar ratios have been carried out at 25 degrees C and at pH 7.0. Ca2+ binding to calmodulin produces heat (-delta H) in the presence of TFP, while heat is absorbed in the absence of TFP. The total heat produced by Ca2+ binding to all four sites is increased at increasing TFP-to-calmodulin ratios, attainin...

The interaction of dihydropyridine calcium channel blockers with calmodulin and calmodulin inhibitors.

The calmodulin inhibitors R24571 and trifluoperazine were found to inhibit competitively the binding of [3H]nitrendipine to a 48,000 X g particulate fraction of rat brain with IC50 values of 1.0 and 18.8 microM, respectively. Equilibrium dialysis was used to test the ability of the dihydropyridines nitrendipine, felodipine, and nicardipine to inhibit the binding of [3H]chlorpromazine, [14C]pimo...
